چکیده

K V 7 (KCNQ) voltage-gated potassium channels are encoded by the KCNQ gene family, many of which display highly restricted and distinct tissue distribution. Cloning experiments have identified 5 KCNQ genes (KCNQ1–5). 1 Mutations in 4 of the 5 genes of the KCNQ gene family have been associated with inherited diseases. KCNQ1 mutations cause cardiac arrhythmia in long QT syndrome with or without deafness. KCNQ2 and KCNQ3 mutations cause benign familial neonatal seizures and peripheral nerve hyperexcit-ability. Mutations in KCNQ4 underlie congenital deafness and modulate cutaneous touch sensitivity in man.

Downregulation of Kv7.4 channel activity in primary and secondary hypertension.

BACKGROUND Voltage-gated potassium (K(+)) channels encoded by KCNQ genes (Kv7 channels) have been identified in various rodent and human blood vessels as key regulators of vascular tone; however, nothing is known about the functional impact of these channels in vascular disease. We ascertained the effect of 3 structurally different activators of Kv7.2 through Kv7.5 channels (BMS-204352, S-1, an...

Contribution of Kv7 channels to basal coronary flow and active response to ischemia.

The goal of the present study was to determine the role of KCNQ-encoded Kv channels (Kv7 channels) in the passive and active regulation of coronary flow in normotensive and hypertensive rats. In left anterior descending coronary arteries from normotensive rats, structurally different Kv7.2 to 7.5 activators produced relaxations, which were considerably less in arteries from hypertensive rats an...

Coupling of L-type Ca2+ channels to KV7/KCNQ channels creates a novel, activity-dependent, homeostatic intrinsic plasticity.

Experience-dependent modification in the electrical properties of central neurons is a form of intrinsic plasticity that occurs during development and has been observed following behavioral learning. We report a novel form of intrinsic plasticity in hippocampal CA1 pyramidal neurons mediated by the KV7/KCNQ and CaV1/L-type Ca2+ channels. Enhancing Ca2+ influx with a conditioning spike train (30...

Small Molecule Investigation of KCNQ Potassium Channels: A Dissertation

Voltage-gated K channels associate with multiple regulatory proteins to form complexes with diverse gating properties and pharmacological sensitivities. Small molecules which activate or inhibit channel function are valuable tools for dissecting the assembly and function of these macromolecular complexes. My thesis focuses on the discovery and use of small molecules to probe the structure and f...
